dc.description.abstract | [SPA] El cálculo del tratamiento térmico que ha recibido un alimento es un punto crítico en cuanto a seguridad alimentaria se refiere. Los microorganismos son excelentes indicadores del nivel de letalidad alcanzado tras un proceso de esterilización. La evaluación microbiológica requiere la utilización de integradores tiempo-temperatura (ITTs), especialmente diseñados para que los esporos de los microorganismos queden inmovilizados, que puedan procesarse y que, analizados posteriormente, permitan comprobar el impacto del calor sobre el alimento. En esta tesis doctoral se han desarrollado dos tipos de ITTs microbiológicos capaces de evaluar la severidad del tratamiento térmico aplicado. Un ITT para la validación de tratamientos térmicos aplicados a alimentos ácidos, seleccionando Alicyclobacillus acidoterrestris como elemento sensor. Para la validación de tratamientos de esterilización de alimentos de baja acidez se desarrolló un ITT con Bacillus sporothermodurans como elemento sensor. A los microorganismos seleccionados se les determinó su termorresistencia bajo condiciones isotérmicas y no isotérmicas simulando los tratamientos térmicos industriales de esterilización de vesículas de mandarina y sopa de verduras. Se obtuvo un valor de D105ºC = 0,63 min y un z = 10,8 ºC en zumo de mandarina para A. acidoterrestris, mostrando curvas de supervivencia lineales, sin hombros ni colas. Para B. sporothermodurans se obtuvieron curvas de supervivencia con hombros que, no obstante, fueron reproducibles y predecibles mediante el uso de modelos no lineales como el de Weibull (valores ?135ºC = 0,13 min y z = 7,6ºC en solución de almidón) o el de Geeraerd (valores D135ºC = 0,10 min y z = 7,9ºC en el mismo medio). Para ambos microorganismos, la predicción de inactivación microbiana bajo condiciones no isotérmicas se pudo realizar adecuadamente a partir de los datos obtenidos en condiciones isotérmicas. Los esporos de los microorganismos seleccionados fueron incluidos en ITTs de alginato y utilizados para evaluar los tratamientos térmicos aplicados en el termorresistómetro Mastia y en un reactor prototipo piloto. Estos tratamientos simulaban los tratamientos térmicos industriales de esterilización de las vesículas de mandarina y sopa de verduras. En ambos equipos los ITTs predijeron con exactitud la letalidad de los tratamientos térmicos aplicados, mostrando que la distribución del calor es homogénea. También permitieron detectar sobreprocesados en los tratamientos aplicados en el reactor prototipo piloto, poniendo de manifiesto la conveniencia de llevar a cabo validaciones biológicas de los tratamientos térmicos que se aplican en la industria alimentaria, especialmente cuando la medida de la temperatura en el interior de los equipos es incierta. | es_ES |
dc.description.abstract | [ENG] The calculation of the heat treatment that a food has received is a critical point in terms of its safety. Microorganisms are excellent indicators of the lethality levels achieved after a sterilization process. The microbiological evaluation requires the use of time-temperature integrators (TTIs), specially designed for microbial spores to be immobilized, that, once processed and subsequently analysed, will allow to check the impact of heat on the product sterility. Two types of microbiological TTIs able to assess the severity of the heat treatment applied were developed in this thesis. A TTI for the validation of thermal treatments applied to acid foods used, selecting Alicyclobacillus acidoterrestris as a sensor element. The other TTI was for low acid foods. In this case Bacillus sporothermodurans was chosen as a sensor element. The heat resistance of these microorganisms was analysed under isothermal and no isothermal conditions simulating the industrial heat treatments of tangerine vesicles and vegetable soup. The heat resistance of A. acidoterrestris was characterized by D105= 0,63 min and z = 10,8 ºC in tangerine juice, showing linear survival curves, without shoulders and tails. The isothermal survival curves of B. sporothermodurans showed shoulders. These curves could be appropriately described by the non linear models of Weibull (δ135ºC = 0,13 min and z = 7,6ºC in starch solution) and Geeraerd (D135ºC = 0,10 min and z = 7,9ºC in this same medium). Predictions of microbial inactivation under non-isothermal conditions was, for both microorganisms, accurately performed from isothermal data. The spores were included in alginate TTIs and they were used to estimate the severity of thermal treatments applied both in a thermoresistometer Mastia and in a food pilot plant scale system. These treatments simulated the sterilisation treatments applied in the food industry for tangerine vesicles and vegetable soups. In both equipments, the TTIs accurately predicted the lethality of the thermal treatments applied, showing that heat distribution was homogeneous. They also enabled to find out overprocessing in the thermal treatments applied in the pilot plant scale system, showing the convenience of performing biological validation of the thermal treatments applied in the food industry, especially when uncertainty in temperature measurements inside the equipments exists. | en |
